Do these syndromes share immunological phenotypes? Last but not least, we try to identify gaps in our knowledge that need further study.Ī systematic search via the online database PubMed of publications on immunological aspects of CHARGE syndrome was performed ( Supplementary 1). We also explore what can be learned from the clinical overlap between CHARGE syndrome and other multiple congenital anomaly (MCA) syndromes. Here, we provide an overview of the current literature on immunological dysfunction in CHARGE syndrome. 8, 9, 13 Furthermore, deficits in the immune system have been described in CHARGE patients, which might lead to morbidity and even mortality. In addition to the above symptoms, other common clinical features of CHARGE syndrome are: absent or hypoplastic semicircular canals, cranial nerve dysfunction (including facial nerve palsy), cleft lip and/or palate, anosmia, feeding difficulties and skeletal abnormalities. Since the genetic cause of CHARGE syndrome was identified, its phenotype has been further explored. There is no clear genotype–phenotype correlation, but variants leading to a premature stop codon are, in general, associated with a more severe phenotype than variants with a non-truncating effect, that is, missense variants. Haploinsufficiency of CHD7 affects multiple organ systems, including the heart, the inner ear and the eye. 6, 9, 10, 11 The CHD7 gene encodes a member of the chromodomain helicase DNA-binding protein family that regulates the transcription of genes during embryonic development. A variant in CHD7 can be found in over 90% of all children who fulfil the clinical diagnostic criteria. 8 Since 2004, when the major genetic cause of CHARGE syndrome was identified as a dominant variant in the CHD7 gene (MIM# 608892), which usually occurs de novo, CHARGE syndrome can also be diagnosed by molecular diagnostics. The incidence of CHARGE syndrome (MIM# 214800) is estimated at 1 in 15 000–17 000 newborns 6 and its clinical diagnosis is based on the criteria proposed by Blake et al 7 or by Verloes. Then in 1981, Pagon et al 5 proposed the acronym CHARGE, which stands for the following symptoms: Coloboma of the eye, Heart defects, Atresia of the choanae, Retardation of growth and/or development, Genital abnormalities, and Ear abnormalities (external, middle and inner ear including deafness). 1, 2 However, the association between coloboma, congenital heart defects and choanal atresia was first recognized by both Hall 3 and Hittner et al 4 in 1979. Patients with a combination of coloboma and other multiple congenital abnormalities were first described in 1961.

Since there’s no published research that’s been performed on this molecule, we dig into the underlying chemistry in the section below. It’s definitely different than anything with PEA or DMAA that we’ve taken in the past – and we’ll explain why below – so it’s got us thinking… if this is FDA compliant, is N-phenethyldimethylamine (or possibly any supplement that has Eria Jarensis Ames on the label) what we’ve been looking for? It’s not over the top, but it’s a more uplifting feel than any fat burner we’ve tried since the original OxyELITE Pro, which had the now-banned DMAA in it, and is more expensive and harder to come by. The best word to describe the feeling is one of euphoria. On Dexamine Black’s label, an interesting new ingredient: N-phenethyldimethylamine, which seemed like yet another PEA-based ingredient.Īlpha Lion SuperHuman Supreme is one of the most potent supplements with this advanced form of PEAīut when trying the Dexamine Black (which is also loaded with a potent 250mg of caffeine – in one capsule), the “buzz” definitely lasted a lot longer. It wasn’t insane, but we could feel that extra little “it” factor so many supplement users seek.Ĭould it be… the next big thing is here? Introducing N-phenethyl dimethylamine, a constituent of Eria Jarensis Ames Something was definitely different with this product. That’s when Giant Sports released the label of now-discontinued “Dexamine Black”, and sent us a couple of early release bottles. But it does pose problems that we’ll get into later.Īnyway, it was back to the stimulant drawing board for the industry, and that’s where we were – up until about two months ago. This is not uncommon – after all, your vitamin C supplement is likely synthetically created, not from freshly squeezed oranges. There is one thing in common with all of these ingredients: in all reality, most manufacturers were most likely just including synthetically-created components, not actually using extract. Hi-Tech Pharma is fighting those letters. It was labeled as a constituent of the much heralded acacia rigidula plant, but the FDA states that they cannot find it in acacia rigidula (nor was it listed on many acacia-based product labels that actually contained it), so they’ve sent warning letters. Meanwhile throughout much of this time, an ingredient known as BMPEA has been on the market. The FDA acted fast, recently putting the kibosh on that one too. Then, a flash in the pan: AMP Citrate, which was supposedly found in the vapors of pouchong tea, provided a nice “DMAA Lite”-like edge. At 250mg caffeine per capsule and an open formula, it isn’t pulling any punches The first supplement with this hot new ingredient. Companies struggled to mix various ingredients and ever-higher doses of caffeine to get that extra edge… but it just wasn’t the same. Īfter that debacle, there was a bit of a layoff, and the industry took a collective sigh, attempting to recover from the negative press whirlwind that affected the entire industry. The first thing after DMAA was Dendrobium extract, which was all the “craze”… until everyone found out that their favorite new pre workout was actually spiked with a methamphetamine analogue, and dendrobium contained none of the PEA-based compounds it was marketed to have. Things got serious, as new regulatory battles (and extremely questionable industry behavior) raged with higher and higher frequency. Once DMAA was banned, the game was on to find the next big thing. Reintroduced by Patrick Arnold and made famous in Jack3d and OxyELITE Pro, it was supposedly found in geranium stems, but never approved by the FDA for use in supplements. NO-Xplode then ruled the streets for a while, but the industry was then completely disrupted when DMAA entered the scene. It all started with ephedrine / ephedra, which the FDA prohibited for use in dietary supplements in 2004, which was finalized in 2006 after a long and brutal legal battle.

Use this calculator to estimate your car payments if you roll your negative equity into a new loan. These calculators are not exact, but can, at the very least, provide you with an estimated figure simply based on your state and estimated purchase price. If your state's DMV website does not provide an online calculator, you can find one on many car-related websites. (Even DMV officials have difficulty in determining exact registration fees, thus the name "estimator." ) The process is easy, requiring entering pertinent information like vehicle identification number (VIN), purchase amount, and license plate type. Given the complexity in determining vehicle registration fees, it's best to use an online calculator, or what some state's refer to as a Fee Estimator. You can do this on your own, or use an online tax calculator. Once you have the tax rate, multiply it with the vehicle's purchase price. Dealership employees are more in tune to tax rates than most government officials. If you are unsure, call any local car dealership and ask for the tax rate. It's fairly simple to calculate, provided you know your region's sales tax. Vehicle tax or sales tax, is based on the vehicle's net purchase price. Realizing the difficulty used car buyers face in properly assessing registration and tax costs for used vehicles, many independent and Department of Motor Vehicle (DMV) websites now provide online vehicle tax and tags calculators. How much will your monthly car payment be Before taking the leap, be sure to understand how much car you can afford each month. New York, for example, bases the fee on a vehicle's weight, while Colorado employs a complicated formula based on the vehicle's year, weight, taxable value, and date of purchase. Appointments & Locations Vehicle registration Learn how to transfer ownership of a vehicle, transfer a title, get tabs on your new vehicle, and change your name or address on a vehicle or boat. Roll capability is new for a Progress launch, provided by the digital guidance system of the Soyuz. Liftoff is set for precisely 16:23:58 UTC as Soyuz rises from its pad to climb vertically before beginning to pitch and roll onto its precise ascent trajectory to begin heading to orbit. Twenty seconds before launch, the RD-107A and RD-108A engines on the boosters and Core Stage come to life and soar up to intermediate thrust levels before throttling up to a total liftoff thrust of 422,000 Kilograms. Heading into the terminal countdown sequence six minutes before liftoff, the Soyuz completes final reconfigurations including propellant tank pressurization, engine purge, and the transfer to onboard power and control. The Guidance System of the rocket will be configured for launch and teams will evacuate the launch pad ahead of the Terminal Countdown Sequence. Fueling wraps up about two hours before launch, allowing teams to perform final close-outs of the launch vehicle and pad structures.ĭuring the final hour of the countdown, the Soyuz will go through a final set of tests and the Service Structure is retracted to clear the way for liftoff. In addition, the boosters and core stage are loaded with liquid Nitrogen for tank pressurization and Hydrogen Peroxide to drive the turbopumps of the engines. The 12 tanks of the Soyuz will be filled with a total of 272,140 Kilograms of rocket-grade Kerosene and –183☌ Liquid Oxygen. After the tanking cars pull up to the pad, they are connected to propellant systems to initiate the propellant loading procedure approximately four hours before liftoff. Five hours before launch, the Russian State Commission convenes to review the status of launch preparations as well as the results of launch vehicle testing to provide the formal approval for propellant loading.Īt that point, teams will be busy working on the Soyuz as final hands-on tasks and fueling preparations are being made. Completing final hands-on work on the launch vehicle, engineers will install batteries in the booster and remove protective covers from the Soyuz including the first stage engine covers.įive hours before launch, the Russian State Commission convenes to review the status of launch preparations as well as the results of launch vehicle testing to provide the formal approval for propellant loading. Communication checks, electrical testing and propulsion system testing is performed in the early stages of the countdown. Countdown & Launch Sequence Photo: RoscosmosĬountdown operations kicked off eight hours ahead of the planned launch time with the activation of the Soyuz launcher and Progress spacecraft for a last round of checkouts and reconfigurations. Progress M-29M entered an independent orbit just below ISS to complete nine days of engineering testing ahead of re-entry. The way is clear for the docking of the Progress to Zvezda’s aft port after it was vacated on Wednesday by the final Progress M spacecraft. Progress MS-02 is loaded with 2,425 Kilograms of cargo for ISS comprised of 1,418kg of dry cargo in the pressurized cargo module of the spacecraft, 540 Kilograms of refueling propellant to be transferred to ISS, 420 Kilograms of water for crew consumption and oxygen generation, and 47 Kilograms of pressurized oxygen and air to top up the Station’s atmosphere. SpaceX is targeting April 8 for the return to flight of the company’s Dragon spacecraft to become the third vehicle to arrive at ISS in a period of three weeks. The first Progress MS flew in December and completed a flawless rendezvous with ISS.ĭocking is set for 18:00 UTC on Saturday to mark the second cargo craft arrival at ISS in just one week in a very busy period of visiting vehicle operations that kicked off back on March 18/19 with the arrival of Soyuz TMA-20M and the Expedition 47/48 crew members, Alexey Ovchinin, Oleg Skripochka and Jeff Williams. This is the last Progress MS flight before the MS upgrade is introduced on the crewed Soyuz in June and teams desire to complete detailed checks of the new systems before flying them on a crewed craft. Progress MS-02, the second in the improved MS series of spacecraft, is set for a two-day chase of the Space Station, taking the scenic route to enable engineers on the ground to put the spacecraft and its various new systems through testing while en-route to ISS. Loaded with over 2,400 Kilograms of cargo, Progress MS-02 is aiming for liftoff on Thursday at 16:23:58 UTC to kick off a nine-minute climb into orbit. Countdown operations are underway at the Baikonur Cosmodrome for a nighttime liftoff of a Soyuz 2-1A rocket carrying the Progress MS-02 spacecraft into orbit for a two-day trip to the International Space Station. When we have friends over for carnitas, I like to make a big batch of this margarita recipe in a pitcher. You can also double or triple this Italian Margarita recipe, rather than making one at a time. I like to garnish with these dried orange slices, or add in a wedge of lime. Shake well then strain into a glass filled with ice.
Constructs from the existing literature can be used to name these categories, particularly if the goal of the research is to extend current theories. Categorization can be done is phases, by combining concepts into subcategories, and then subcategories into higher order categories. Categories are needed to reduce the amount of concepts the researcher must work with and to build a “big picture” of the issues salient to understanding a social phenomenon. While concepts may be context-specific, categories tend to be broad and generalizable, and ultimately evolve into constructs in a grounded theory. Next, similar concepts are grouped into higher order categories. This coding technique is called “open” because the researcher is open to and actively seeking new concepts relevant to the phenomenon of interest. While coding, it is important to identify the recognizable characteristics of each concept, such as its size, color, or level (e.g., high or low), so that similar concepts can be grouped together later. Once a basic set of concepts are identified, these concepts can then be used to code the remainder of the data, while simultaneously looking for new concepts and refining old concepts. Concepts can be named using the researcher’s own naming convention or standardized labels taken from the research literature. Simple concepts such as “organizational size” may include just a few words of text, while complex ones such as “organizational mission” may span several pages. The coding unit may vary with the concepts being extracted. Some concepts may be simple, clear, and unambiguous while others may be complex, ambiguous, and viewed differently by different participants. Each concept is linked to specific portions of the text (coding unit) for later validation. The researcher examines the raw textual data line by line to identify discrete events, incidents, ideas, actions, perceptions, and interactions of relevance that are coded as concepts (hence called in vivo codes ). Open coding is a process aimed at identifying concepts or key ideas that are hidden within textual data, which are potentially related to the phenomenon of interest. Strauss and Corbin (1998) describe three coding techniques for analyzing text data: open, axial, and selective. To ensure that the theory is based solely on observed evidence, the grounded theory approach requires that researchers suspend any preexisting theoretical expectations or biases before data analysis, and let the data dictate the formulation of the theory. The interpretations are “grounded in” (or based on) observed empirical data, hence the name. The technique was developed by Glaser and Strauss (1967) in their method of constant comparative analysis of grounded theory research, and further refined by Strauss and Corbin (1990) to further illustrate specific coding techniques – a process of classifying and categorizing text data segments into a set of codes (concepts), categories (constructs), and relationships. How can you analyze a vast set qualitative data acquired through participant observation, in-depth interviews, focus groups, narratives of audio/video recordings, or secondary documents? One of these techniques for analyzing text data is grounded theory – an inductive technique of interpreting recorded data about a social phenomenon to build theories about that phenomenon. Interested readers are referred to more authoritative and detailed references such as Miles and Huberman’s (1984) seminal book on this topic. This chapter provides a brief overview of some of these qualitative analysis strategies. A creative and investigative mindset is needed for qualitative analysis, based on an ethically enlightened and participant-in-context attitude, and a set of analytic strategies. The emphasis in qualitative analysis is “sense making” or understanding a phenomenon, rather than predicting or explaining. Unlike quantitative analysis, which is statistics driven and largely independent of the researcher, qualitative analysis is heavily dependent on the researcher’s analytic and integrative skills and personal knowledge of the social context where the data is collected. Qualitative analysis is the analysis of qualitative data such as text data from interview transcripts. Older kids love the complicated, intricate and challenging designs.
